Abstract

The commonly used media for microbiological examination is Nutrient Agar as a general media, but the
media price is expensive. Tofu dregs is one of the natural ingredients that contain protein is high enough and
the price is cheap while the waste has not been optimally utilized. This study aims to determine the
characteristics and number of colonies of Bacillus sp. on media waste of tofu dregs. This research was
conducted by using experiment laboratory method that is looking at the growth of Bacillus sp. on alternative
media from waste of tofu dregs with variation of concentration that is 6%, 7%, 8%, 9% and 10% with double
repetition technique (duplo). Based on the result of research, characteristic of colony Bacillus sp. on medium
Nutrient Agar is round, yellowish white, diameter 1-2 mm, flat elevation, rough surface, and uneven edges of
colony. Whereas in the medium waste of tofu dregs that is round, white, has a small diameter, the edge of the
colony widens and looks like a transparent fiber, flat elevation and rough surface. The total colony on the
media Nutrient Agar and media waste of tofu dregs at concentrations of 6%, 7%, 8%, 9%, 10% respectively,
